After the death of their father, the Elric brothers, along with their mother and younger brother Roy, are exiled by the country of Amestris, where they live in an isolated village known as Roy's Palette. They continue to practice alchemy, hoping to find a way to restore their bodies. After narrowly escaping the attack of the homunculi, the brothers reunite with their half-sister, Winry Rockbell, and come across a half-mad young woman named Pinako. The siblings later find their mother's kidnapped sister, Rebecca. When they learn that Rebecca is an adopted daughter of the Homunculi, they realize that she is in serious danger, and that they have only a few days to find a way to save her. Together, the siblings, along with their allies Nina and Marshall, set out in search of the legendary Philosopher's Stone, a powerful alchemical substance that can grant an alchemist any wish. Along the way, they encounter the elements of air, fire, earth, and water that they face, and come to terms with their new spiritual powers.
